Abstract
PRe-ART (Predictive Reagent Antibody Replacement Technology) aims to replace animal-derived, underperforming reagent antibodies with designed armadillo repeat
proteins (dArmRPs) for widespread use as protein-binding reagents, in biomedical research. These rationally designed, modular proteins bind linear target peptides in a conserved and extended conformation. Based on a designed and highly stable armadillo repeat protein, individual armadillo repeats are engineered by a combination of computational design, high-throughput mutagenesis and high throughput affinity screening to achieve single amino acid specificity. Variant repeats are then assembled according to the required target in a modular manner, ultimately to bind protein/peptide targets on demand.
